The Opentrons Flex Automated Vacuum Module GEN1 brings vacuum-based filtration into fully automated workflows on the Opentrons Flex. Using a software-controlled pump and gripper-compatible manifold, the module enables plate-based vacuum filtration without manual pressure adjustment, mid-protocol pauses, or off-deck equipment.
Common applications include nucleic acid extraction and miniprep workflows, protein and peptide desalting before LC-MS, and solid phase extraction (SPE) for proteomics and small molecule sample prep. The module supports both filter-to-waste and filter-to-collection-plate configurations, and concurrent pipetting is available while vacuum is applied.
The module ships as a complete, easy-to-set-up kit: manifold base, short and tall collars, spacers, 2L waste carboy, pump, and all tubing and cabling. It occupies two deck positions (A3 and A4) and connects to the Opentrons Flex via USB. Setup, calibration, and vacuum control are all managed through the Opentrons App and standard protocol commands.

Filtered waste routes through the manifold base directly into the included 2L carboy. The system includes a binary liquid-level sensor that triggers an error recovery prompt before the carboy reaches capacity. At protocol start, the app will remind you to confirm the carboy is empty.
The module is designed around standard SBS-footprint 96-well filter plates operating in the -270 to -800 mbar gauge (atmospheric to 200mbar atmospheric) range.
